Ian, I had no issues with turning on File Vault while using Voiceover. Everything is straight forward. You must set a master password to unlock the computer and decrypt your files if you forget your login password. If you have a large home folder, the encryption does take a while. I have about 230 GB in mine. I left my iMac on overnight as it did the encryption.
As your files are being encrypted, you cannot use your Mac, so I'd recommend turning File Vault on before bed and letting it do its thing.
