The VoiceOver drag and drop action that you need to use may be buggy, so read down the thread with the Control-N access key combination to see Donna's reply and Anne's comment. There's an old thread where signatures in mail was discussed, along with the issue of drag and drop. Anne's post is more succinct and up to date, but if you want to read the old thread from last year about creating a signature in mail for background, it's at:

Again, you'll want to use the access key combination Control-N to read the next post in the thread (or Control-P to move back to previous post) to read through to the discussion about the drag and drop issue.

Another way to get around this drag and drop business of setting up signatures would be to use Snow Leopard's new text expansion features. This is described in an article titled "Using Snow Leopard's built-in text snippets in Mail.app":

This basically describes a way to select a "Text Replacement" sub-menu option under the "Substitutions" option under Edit in mail to define a string to be used for some predefined key of your choosing. For example, you could decide to have a phrase like "ssig1" to get expanded into the lines for your name and home address (which you typed into TextEdit, and then copied and pasted into the replacement field). Then, whenever you typed that phrase in mail, it would expand to your replacement definition (and you could use it anywhere in mail, not just in the signature line). You're basically using the System Preferences > Language & Text menu features under the Text pane to set up these replacement strings, but the steps described in the article working from the Edit menu in Mail will take you to these options. This article has a good description of other text replacement features under Snow Leopard, and is worth reading.
