Hi Michael, I'm using LyX 2beta3 on OS X 10.6.6 with MacTeX also. I have compiled a document with the tufte class both to DVI and to PDF. This suggests that there is something specifically wrong with your file. It could be your installation, but if you used the full MacTeX, that seems doubtful.
If you can post your actual minimal sample document for those who are the experts (among whom I can't be counted), someone might be more able to give a better analysis of the specific LyX/LaTeX item that is causing the trouble.
