This means that LyX cannot find Python. Have you installed Python manually previously? If not, LyX didn't automatically install it due to registry settings that might remain from earlier installations of Python. In this case do:

- check the registry that there are no entries saying that Python is installed
- uninstall LyX _completely_ (but don't uninstall MiKTeX)
- open an Internet connection
- reinstall LyX using the small variant of this installer:
  http://wiki.lyx.org/Windows/LyXWinInstaller
