In LyX 1.2.0 it is easy to do (the insert reference dialog allow listing the labels in other buffers). In Lyx <= 1.1.6, perhaps the simplest way is to go the the 2nd chapter, insert the reference there, and then cut the reference, return to the original chapter, and paste the reference.
