It is a way but the problem I find is that the file won't compile with
that commandline option.
Say your file is
% this is example.ly
#(use-modules (guile-user))
#(set-global-staff-size size)
\repeat unfold 20 { c'1 }
%END
you can call it with
lilypond -e '(define-public size 40)' a.ly
and the value of size will be honoured as global staff size.
