You can use both products altogether, but only one can be configured for 
real-time protection.
Windows now takes care of denying activation of WD real-time protection if 
you're using a third party AV like VIPRE. You can, in this case, use your third 
party AV product for real-time protection and configure Windows Defender to 
perform periodic scanning at regular intervals.
Depending on the Windows 10 build, you can configure this on Windows Defender 
Security Center or, in older builds, the Update and Security\Windows Defender 
category in the Settings app (WINDOWS+I).
