To start the agent you need to add the --daemon argument. For testing you may use this: gpg-agent --use-standard --daemon sh which opens a new shell and sets up everything. You need to make sure that no other agent is running and controlling the card. You should also unset the GPG_AGENT_INFO ebvar which might have been set by another script.
