There isn't really anything Django-specific here, although there might be helper apps. But take a look at FlowPlayer, which is a free, customisable Flash video player (I think it plays both FLV and H.264). For converting videos, ffmpeg is the thing, and a quick Google suggests there are at least a couple of Python libraries that use the ffmpeg libraries: PyMedia and pyffmpeg.
