Not sure if this is a bug or expected behavior for Nautilus now. Can confirm that the middle-mouse button does open a file with a single click. Both the left or middle mouse buttons can drag and drop a file by moving the file, not copying the file. No option appears to select which method to use.

In Ubuntu 16.04 I could middle click on a file in Nautilus then drag and drop it to another folder. After that a small dialog appeared if I want to copy or move the file. Now when I press middle click on a file, the file opens immediately and although I can move it to another folder, there's no choice if I want to copy or move the file.
