I take it this isn't about using DOSMU but a virtual machine with a FAT16 file system like going back to 1987. How can I do that?

You can install virtualbox for example. Within the program you create a harddrive and configure the parameter for the virtual machine (memory, hardware). When you launch the VM, you can install DOS/Windows/Linux like you do on normal hardware.

If you have floppy or CD-ROM, you can allow the VM to access these devices directly. Or you can create images of the software and attach the image to the VM.
