What payers are you using? Try a low level alsa tool, like aplay. BTW, alsa is known for muting the mixer by default. Did you check this? Did you do any changes to your hardware? Does sound work with a live-cd? In your OP I can see you mention you needed to use alsaconf after an upgrade, but then even this didn't work. What did you change?
