The .dsc file is the Debian Source Control file. It specifies which components are part of the Debian source package. To create a new Debian package you can read the New Maintainer guide and create a proper Debian package, or you can install the checkinstall package and use it to "create" a Debian package, in the it will register a package with dpkg that lists all the files installed in the 'make install' process of the software.
