If you have at least 1GB of RAM, you can boot Knoppix with the option "toram", this will allow you to remove the CD. To do this with less RAM, you would need to shrink the CD size by remastering it with some applications removed. Or you can use the option "tohd=/dev/hda1" (adjust device as needed) if you don't mind touching the contents of your harddisk.
