This is a team maintained package, and normally it should be a plain "GBP" style layout.
Looks like in 2013 "single-debian-patch" was added to debian/source/options by Charles Plessy: https://salsa.debian.org/med-team/htslib/-/blame/master/debian/source/options?ref_type=heads#L1 However I did a test build locally and that wasn't needed, so I'll probably remove that option unless someone informs me otherwise.
