img2pdf now refuses to work on input that it cannot process without loss of information. PDF is not able to retain transparency information. That's why img2pdf refuses to work on input that contains an alpha channel. If you really want to process an image that has an alpha channel, then you have to remove it yourself before passing the image to img2pdf. This is a change that ocrmypdf has to implement.
