That purely depends on what you're going to put into those columns. If you're putting images or a fixed width google ad in there, then they should be sized in pixels, to discourage breakage at smaller window sizes. If you do decide to make the columns fluid, I'd also consider setting a min width and a max width on the layout, since narrowing the window a lot may make the columns break out at smaller resolutions with bigger default font sizes.
