Use butyl tape as sealant. 2. Be aware that hull and deck will flex open differently when removing the screws. So do 1/2 or 1/3 of the length each session. If the boat will be floating, the flexing will be dynamically active with the waves/surf. You need cover from rain. 3. Study about tensioning studs technics online, it is not as easy as tightening... Studs and nuts cannot be reused they get deformed while tensioning ( in two steps), use of washers, this is a deep subject.
