Best, Ingrid The groups of Ingrid Span at the Institut für Physikalische Biologie and Ursula Fleig at the Institut für Funktionelle Genomforschung der Mikroorganismen at Heinrich-Heine-Universität Düsseldorf are seeking to fill a PhD position in structural biology and cell biological research (f/m/d) within the Graduate Program for Biology Science (iGRADbio) at HHU Düsseldorf. The successful candidate for this position will focus on studying the structure and molecular mechanisms of the iron-sulfur protein Asp1, a member of the Vip1/PPIP5K enzyme family. This enzyme family was discovered 13 years ago and is a highly conserved, exclusively eukaryotic enzyme family that controls the intracellular levels of a signaling molecule named inositol pyrophosphate (IPPs). Since then the importance of this Vip1/PPIP5K enzyme family in numerous cellular processes has become evident. Among other things, Vip1/PPIP5K made-IPPs control hearing in humans, mammalian organ development, the response to pathogen attack in mammals and plants and the faithful inheritance of chromosomes in fungi. Vip1/PPIP5K proteins are bifunctional enzymes comprised of autonomous kinase and pyrophosphatase domains that manufacture or destroy specific IPPs, respectively. How the two enzymatic activities are controlled on a molecular level is unclear and the focus of this PhD thesis. The project involves X-ray crystallography in combination with genetic, cell biological, biochemical, and spectroscopic methods. Candidates should have a strong background in biology and/or chemistry. Prior research experience in X-ray crystallography or metalloproteins or molecular cell biology are desirable but not required. Candidates should also have strong communication skills, the ability to conduct research, and the ability and willingness to work in an interdisciplinary team.
