Seungil Han www.pfizercareers.com Postdoctoral Fellow-Structural Biology Job ID: 1042552 Location: United States-Connecticut-Groton Role Summary The successful candidate will join the department of Structural and Molecular Sciences (SMS), a multidisciplinary group with established expertise in molecular biology, protein biochemistry, protein mass spectrometry, enzymology, NMR, biophysics, protein crystallography and cryo-EM. Our mission is to provide integrated molecular insights into drug target mechanism and target-ligand interactions to promote discovery efforts across multiple therapeutic areas, including Inflammation and Immunology, Neuroscience, Cardiovascular, Metabolic & Endocrine Diseases, Pain, Rare Diseases and Oncology. Within Pfizer, we have made a strong commitment to invest in single particle cryo-EM and have set up a state of the art microscope (Titan Krios) with a K2 direct electron detector in newly renovated laboratory, dedicated computer cluster, GPU box and a TF20 microscope for screening samples. As a Postdoctoral Fellow, you will have training & experience with structure determination by cryo-EM technology including sample preparation, data collection, data processing using state-of-art software, and 3-D reconstruction. Ideally you will be familiar with protein construct design, molecular biology and mutagenesis, protein expression, purification and biochemical/biophysical characterization of a membrane protein. Equally important will be your ability to collaborate successfully with other scientists in the department and with biologists and chemists in multiple drug discovery teams. You will eventually be expected to understand the critical issues and challenges faced by different drug discovery programs within Pfizer and design suitable experimental strategies based on cryo-EM technique that could positively impact the outcomes. SMS is part of Medicine Design group within Pfizer, with the overall goal of understanding the mode of action of lead compound using an integrated assessment of all aspects of protein-ligand interactions to drive drug discovery efforts. Role Responsibilities The Postdoctoral Fellow will design and perform experiments in the technical area of membrane protein purification, characterization and involved in single particle cryo-EM. Experimental results will be formulated computationally and will be recorded in an electronic laboratory notebook. Prompt and clear communication will occur with project team colleagues. External scientific publication and presentation will be encouraged when appropriate as a means of assuring optimal research standards. Qualifications • A PhD or be a PhD candidate (to be completed in 2016) in Biochemistry, Biophysics, Physics, Chemistry or a related field. • Desire to learn and carry out single particle cryo-EM work with suitable independence, and have the creativity and drive to tackle scientific challenges.
