Dear CCP4 members I do not have any good experience with chemical cross linking, it has not worked for me and I am curious how others have got it to work. In my latest attempt I wished to crosslink two monomers with DSS. The protein is exclusively dimeric according to SEC. I incubated the protein with a 20 times molar excess of the DSS for 1 hour at room temperature but when I ran it on a gel, all lanes look like the control, ie the crosslinking was completely unsuccessful.
I wonder if any of you have any good experience advice or tips for me? I used DSS as it was used to cross link a similar protein, its spacer distance is around 11A. Best Careina.
