Dear Amr, Every protein is different, so one cannot make a general remark. However, in my experience disulfide bonds are normally not affected by mercaptoethanol or DTT. I have had one protein where the disulfide bonds were not made, likely because the protein was produced in E.coli. However, the protein still crystallized. Good luck! Herman
