Hi marija, you could try mass-spec. It shouldn't be too hard if your protein is a soluble protein (membrane proteins are more complicated). As a side note, we have never checked Semet incorporation and I'm sure this is true for many others. If you use the common protocols (either use an auxotroph or do met biosynthesis in a wildtype strain) there's no reason why you wouldn't get good incorporation.
