Can't help you from the perl angle, but coming from the Exchange side I would hazard a guess that the server is correctly configured to not relay. Is it an option for you to run SMTP (of whatever flavor) locally and call the mail app in a sendmail-like fashion? If you must forward to an SMTP server, Exchange is tough to work with.
