Perhaps your microphone needs +5V bias to be switched on.  Mine does.
Look for this setting in /etc/asound.state - mine reads...

 control.44 {
                comment.access 'read write'
                comment.type BOOLEAN
                iface CARD
                name 'Mic +5V bias'
                value true

Also gamix (a very nice Alsa mixer for X) has a button to enable this.
