As per this guide, the CampaignCriterionService and AdGroupCriterionService support targeting or excluding various types of criteria. Having said that, you may add the negative keywords by using the CampaignCriterionService. Additionally, you may also use the SharedSetService.MutateSharedSets to create negative_keywords shared set too. Note that SharedSets are used for sharing criterion exclusions across multiple campaigns. Let us know if this is what you're looking for.
